MANTLE CELL LYMPHOMA (MCL)

t(11;14) TRANSLOCATION AND OVER-EXPRESSION of CYCLIN D1

Mantle cell lymphoma is characterised by the presence of the t(11;14) translocation which results in over-expression of the BCL-1 gene and consequent overproduction of cyclin D1. The t(11;14) translocation can be detected using PCR in approximately 40% of  cases of mantle cell lymphoma, using primers described by Van Dongen et al (2003) Leukemia 17, 2257–2317).

Over-expression of cyclin D1 can be identified using Real-time PCR (Howe et al. (2004) Clinical Chemistry (50) 1 80-87).

t(11;14) translocation and cyclin D1 analysis can therefore be used as a tool in the differential diagnosis of mantle cell lymphoma. However, all molecular data must be interpreted with other histological and clinical data.
